G protein-coupled receptors (GPCRs) widely exist in the animal and plant kingdoms, and are also the largest family of membrane receptor proteins in the human body. They are widely involved in the regulation of physiological activities such as cell proliferation, differentiation, and migration. The structural characteristics and important role in signal transduction determine that it can be used as a good drug target. It can be used for the screening of drugs for infectious diseases, cardiovascular diseases, neurology, psychiatric diseases, AIDS, diabetes, etc.
Reqbio is a R&D outsourcing service company that provides GPCR-related solutions, providing a complete set of R&D services, including the construction of GPCR stable cell lines, calcium flux, second messengers (cAMP, IP-One), reporter genes, Beta-Arrestin recruitment, etc. and other detection methods. It not only provides customers with high-quality screening model construction services, but also provides high-throughput screening and compound profiling services. Reqbio's extensive experience will provide you with high-quality service and high-quality results.
Service Description
1. Which screening method is suitable for different targets, please contact us in advance to evaluate your experiment;
2. Drugs, cells and other materials involved in the experiment can be provided by the customer, or can be found in our stock product library, or purchased by us;
Reporter Gene
After the drug binds to the GPCR, the GPCR is activated and starts intracellular signal transduction. According to different pathways, it can be divided into Gq, Gs, and Gi. The three pathways can induce the expression of the nuclear reporter gene (luciferase) through different effector elements. , by detecting the signal of luciferase, indirectly expressing the activity of the drug on GPCR.
The capture signal of this system is chemiluminescence, the background signal is low, and the sensitivity is high. It is suitable for most GPCRs. The disadvantage is that a luciferase cell line needs to be established first.
